Research Summary

During my career I have had extensive experience and published several manuscripts/scientific abstracts relating to stem cell production, culture, isolation, differentiation and transplantation (via both cell injection and polymer scaffold mediated delivery). Two studies of particular importance to this proposal pertain to the use of skin-derived primary cells for the purpose of iPSC generation. The first, published in Plos One, was focused on the use of adult mouse skin derived iPSCs for the purpose of photoreceptor cell production and transplantation induced retinal regeneration. The second, published in PNAS, was focused on production of adult retinal disease specific iPSCs and subsequent human photoreceptors for the purpose of disease modeling and elucidation of disease mechanism. In both studies adult skin derived primary cells were essential for iPSC generation and subsequent experimentation. To date we have generated in excess of 300 molecularly confirmed disease specific human fibroblast and keratinocyte cell lines many of which have been reprogrammed to pluripotency and are now being used for disease modeling, drug screening, and cell transplantation studies.
